The probing of yam (Dioscorea sp) DNA with a heterologous yeast phosphofrutokinase gene

DNA, RNA and mRNA were isolated from leaves of Dioscorea alata, Dioscorea bulbifera and tubers of Dioscorea rotundata and probed with heterologous yeast phosphofrukinase cDNA at varied hybridization temperatures and washing conditions. Southern and Northern blot analyses showed that yeast PFK1 gene were codes for the a-subunit of yeast phosphofrukinase, has sufficient homology to yam DNA sequences to allow its use as a probe to screen yam genomic libraries. The results indicate that yam PFK gene sequences may be located in a 21 kb fragment of ECoRI or BamHl digested yam tuber DNA, a 5 kb fragment of an ECoRI digest, and the 3 kb fragment of a BamHl digest, or on yam 3.5 - 4.5 mRNA. It is not clear at present whether yam genome contains a small PFK gene family, or whether the PFK gene, uniquely present in yam, possesses ECoRI and/or BamHl restriction sites.....
