Improved sprouting rates in Dioscorea cayenensis (cv. round leaf) minisetts using biochemically corroborated chemical treatments

Glutathione, b-amylase, o-diphenolase (EC 126.96.36.199) and peroxidase (EC 188.8.131.52) activities were assayed in the head, middle and tail portions of sprouting and dormant Dioscorea alata (cv. sweet yam) and Dioscorea cayenensis (cv. round leaf) tubers. The results revealed that sprouting initiation of both species studied correlated well with increased glutathione levels and lower o-diphenolase activity. There was also a gradual decrease in the levels of the parameters assayed as one moved from the head to the tail region of the tubers. Minisetts prepared from the head, middle and tail sections of D. cayenesis tubers showed improved sprouting and decreased rotting when treated with 1% ascorbic acid.
