Plasma Homocysteine Levels, Folate Intake and Polymorphisms in the Methylenetetrahydrofolate Reductase Gene in Jamaican women

Objective: To determine the prevalence of methylenetetrahydrofolate reductase (MTHFR) mutations in Jamaican women, their association with homocysteine levels and folate intake. Methods: The study was conducted in two parts : (a) 50 women with pre-eclampsia (PE) and 50 controls were recruited from the Antenatal Ward at the University Hospital of the West Indies (UHWI) and (b) 146 women at different stages of pregnancy were chosen randomly from the Antenatal Clinic, UHWI. Blood was collected. Participants were interviewed about their folate intake using a short food frequency questionnaire. Total plasma homocysteine levels were analyzed by HPLC and fluorescence detection. DNA was prepared from buffy coats. Subjects were genotyped for MTHFR polymorphisms at nucleotides 677 (C ® T), 1298 (A ® C) and 1317 (T ® C). Data were analyzed using ANOVA and X2 tests. Conclusion: Homocysteine levels change significantly during the third trimester. MTHFR polymorphisms were not associated with homocysteine levels.
