Plasmid content of Extended Spectrum Betalactamase (ESBL) producing Eschericha coli [Abstract]

Objective: To identify E Coli producing extended-spectrum beta-lactamase (ESBL) and examine plasmid content asssociated with this resistance mechanism.Method: Clinical laboratory isolate from University Hospital of the West Indies patients were examined for resistance to the third generation cephalosporins: ceftazidime and cefotaxime. A double-disc diffusion test was used to confirm the presence of clavulanate-inhibitable ESBLs. Isolates were grown to 20 ml volume in selective broth for plasmid extraction using PlasmidPURE midipreps. DNA obtained was electrophoresed at 12 V/cm through 1.0% agarose in TBE buffer for up to 8 hours and then stained wih ethidium bromide.Conclusion: High molecular weight plasmids conferring ESBL production E coli are expected to have transferred from resident ESBL klebsiella. The result obained is consistent with this theory, butmore sophisticated analysis is required for proof.....
