Isolation and Identification of Pathogenic Acanthamoeba Strains from Water Sources in Jamaica, West Indies [Abstract]

Objective: To determine the presence and prevalence of free-living opportunistic protozoan parasites of the genus Acanthamoeba in tap and sea water sources in Jamaica. Method: In this study, 62 samples were collected from beaches and tap water sources across the island of Jamaica. A litre of each was water sample was filtered through a cellulose nitrate filter (0.45 Ám diameter, Millipore Corporation, Bedford, Massachusettes, USA) with a week vacuum (flow rate, 1.3 ml/min). The filters were inverted on 2% Neff's saline non-nutrient agar plates and incubated at 25oC. These plates were monitored for out-growth of Acanthamoeba microscopically and blocks containing Acanthamoeba were removed and the amoebae cloned by dilution. The identification of Acanthamoeba isolates was characterised by temperature, osomotolerance assays and PCR.....
