Abstract
Objective: To determine whether a single nucleotide polymorphism (SNP) in the promoter region of the pro-inflammatory cytokine TNFa is associated with cervical dysplasia. Methods: Blood was obtained from women with cervical dysplasia at the Colposcopy Clinic (n = 240) and controls from the Gynaecology Clinic (n = 102). Genomic DNA was prepared from buffy coats and subjects genotyped for the SNP, 308 (G ® A). X2 was used to compare frequencies of the different genotypes and logistic regression analysis used to determine odds ratios (ORs) and 95% confidence intervals (CIs) associated with presence of allele A. Conclusions: The results indicate a possible role for this variant in cervical dysplasia. Association may be either due to TNFà's biological effects or through linkage with other MHC haplotypes.....
