Abstract
Objective: To determine whether a single nucleotide polymorphism (SNP) in the promoter region of the pro-inflammatory cytokine TNFa is associated with cervical dysplasia. Methods: Blood was obtained from women with cervical dysplasia at the Colposcopy Clinic (n = 240) and controls from the Gynaecology Clinic (n = 102). Genomic DNA was prepared from buffy coats and subjects genotyped for the SNP, 308 (G A). X2 was used to compare frequencies of the different genotypes and logistic regression analysis used to determine odds ratios (ORs) and 95% confidence intervals (CIs) associated with presence of allele A. Conclusions: The results indicate a possible role for this variant in cervical dysplasia. Association may be either due to TNF's biological effects or through linkage with other MHC haplotypes.....
